Effect of semen extenders on the motility and viability of stored African Catfish (Clarias gariepinus) spermatozoa

چکیده

This study assessed the effects of common extenders and diluents on the spermatozoa of African catfish (Clarias gariepinus), with the intent of obtaining a semen preservation protocol that can serve as a means of making fingerlings available to fish farmers all year round. Semen samples (milt) were pooled from mature broodstock males and pre-extension qualities were evaluated. Egg yolk, tomato juice and sodium citrate buffers were combined in four different trials and used to preserve obtained semen at temperatures of 5°C and -40°C for 8 days and 4 weeks respectively in different trials. Motility indices of extended, preserved, refrigerated and frozen semen were used to analyze the effect of the various combinations. Forward, progressive and rectilinear motion was employed as a significant measurement of the livability of extended sperm cells and the efficacy of the extender solution. The result of the study showed that the buffer (sodium citrate) on its own performed excellently well (sperm motility was 85%) in enhancing survival within the first 24-48 hours post extension; Semen extended with 20% tomato juice gave the best survival rate when stored at 5°C, because the sperm cells were still viable by day 6 postextension. Extender containing 20% egg yolk gave 70% motility while that containing 10% egg yolk mixed with 10% tomato juice gave 60% motility. However, motility decreased progressively as the period of storage increased. The results of the effect of freezing (at -40°C) on motility revealed that no motility was observed in all the cryopreserved trials except the sample containing 10% egg yolk and 10% tomato juice, which recorded 10% post-thaw motility.

P-73: Effect of Sodium Dodecyl Sulfate and Different Levels of Clove Bud Extract on Motility, Viability and Plasma Membrane Integrity of Ram Spermatozoa during Cooling and Cryopreservation

Background: Hen egg yolk is a common component of most semen cryopreservation extenders. However, diluents containing the egg yolk may have adverse effects on the quality of the semen. Egg yolk could be solubilized by the addition of sodium dodecyl sulphate (SDS) in semen extenders. Many studies have shown beneficial effects of antioxidants addition to semen freezing extenders. Antioxidants are...

The effect of antioxidant supplementation in semen extenders on semen quality and lipid peroxidation of chilled bull spermatozoa

The aim of this study was to determine the effects of the antioxidants superoxide dismutase (SOD),glutathione peroxidase (GPx) and butylated hydroxytoluene (BHT) on microscopic semen parameters andlipid peroxidation following the freeze-thawing of bull semen. Ejaculates were collected from five Holsteinbulls and pooled at 37°C. The semen samples were diluted with a CEY extender containing addit...

Induction of triploidy with caffeine treatment in the African catfish (Clarias gariepinus)

Induction of triploidy is one of the biotechnological methods in aquaculture used for genetic manipulation. It refers to a state where organisms have three complete sets of chromosomes instead of two and can result in sterility. Caffeine treatment that is safe and inexpensive, serve to induce triploidy in catfish. To suppress the second meiotic division, fertilized eggs were exposed to three di...

Responses of growth rates and growth hormone levels of African catfish (Clarias gariepinus) to stocking density

In order to estimate responses of growth rate and growth hormone of Clarias gariepinus to stocking density, fish (initial weight 30.71 ± 0.89 g) were stocked in 120-L tanks at densities of 35, 65, 95, and 125 kg/m3 for 60 days. On day 30 and 60, the fish growth rates were measured, pituitary growth hormone (GH) mRNA levels and serum GH concentrations were examined by real-time qPCR and ELISA te...

Effect of various levels of catalase antioxidant in semen extenders on lipid peroxidation and semen quality after the freeze-thawing bull semen

The objective of this study was to evaluate effect of different concentrations of catalase in two extenders on motility, viability and lipid peroxidation bull spermatozoa during semen freezing process. Thirty ejaculates collected from ten Holstein bulls were pooled and evaluated at 37 °C. Pool ejaculated was split into two main experimental groups, 1 and 2. In experiment 1, specimen was dilute...
